GGT activity is determined by a coupled enzyme assay, in which the GGT transfers the γ-glutamyl group from quenched substrate L-γ-Glutamyl-7-amido-4-methylcoumarin (L--Glutamyl-AMC), liberating the fluorescent group, 7-Amino-4-Methyl Coumarin (AMC) (λex = 365 nm/λem = 460 nm) proportional to the GGT activity present. One unit of GGT is the amount of enzyme that will generate 1.0 μmole of AMC per minute at 37 °C.
